Methylphenidate-OROS is a once-daily, extended-release oral formulation of the central nervous system stimulant methylphenidate. It uses the Osmotic Controlled Release Oral Delivery System (OROS) to provide both immediate and sustained release of the active ingredient throughout the day. The OROS tablet consists of an osmotically active trilayer core surrounded by a semipermeable membrane with an immediate-release drug overcoat; 22% of the dose is released immediately and 78% gradually over 6–12 hours[5][7]. Methylphenidate acts primarily by blocking dopamine and norepinephrine reuptake into presynaptic neurons, increasing their levels in synaptic clefts[1][6]. It is indicated for attention deficit hyperactivity disorder (ADHD) in children, adolescents, and adults as well as narcolepsy[2][4][5]. The controlled release profile allows for consistent plasma concentrations and reduces dosing frequency compared to immediate-release formulations.
