Drug intelligence / Profile preview
Osmotic release methylphenidate refers to a specific extended-release formulation of the stimulant medication methylphenidate that uses an Osmotic Release Oral System (OROS) technology. This delivery system provides both an immediate-release and a controlled extended-release phase. Upon ingestion, about 22% of the dose is released immediately for rapid onset of action; the remaining 78% is gradually released over 10–12 hours via an osmotically driven push mechanism through a laser-drilled orifice in the tablet[1][3][4]. The primary indication for this drug is attention-deficit hyperactivity disorder (ADHD) in children and adults[2][4]. Methylphenidate acts mainly as a norepinephrine-dopamine reuptake inhibitor (NDRI), blocking dopamine transporter and norepinephrine transporter to increase synaptic concentrations of these neurotransmitters in the central nervous system[6][8]. The original developer of this technology was ALZA Corporation.
