Metisazone (also known as methisazone) is a thiosemicarbazone-derived antiviral drug historically used for the prophylaxis and treatment of smallpox and complications from smallpox vaccination. Its primary mechanism of action is the inhibition of viral messenger RNA (mRNA) and protein synthesis, particularly in poxviruses such as variola (smallpox virus) and vaccinia. By interfering with these processes, metisazone prevents viral replication within host cells. The drug was mainly used during the 1960s but saw limited adoption due to logistical challenges in mass administration and significant side effects like nausea. It has also been studied for its potential activity against other viruses in vitro, including SARS-CoV-2 through molecular docking studies[2][3][5][7][8]. Metisazone belongs to the chemical class of indoles and derivatives.
