Metixene (also known as methixene) is a small molecule anticholinergic drug used primarily as an antiparkinsonian agent. It acts as a tertiary antimuscarinic with both central and peripheral actions similar to those of atropine. Metixene competitively antagonizes acetylcholine at muscarinic receptors in the corpus striatum, thereby helping to restore the balance between excitatory cholinergic and inhibitory dopaminergic systems implicated in parkinsonism. In addition to its antimuscarinic effects, it also exhibits antihistaminic and direct antispasmodic properties. Metixene was indicated for symptomatic treatment of Parkinson's disease and for alleviating extrapyramidal symptoms induced by other drugs such as phenothiazines; however, it is not effective against tardive dyskinesias. The drug has been discontinued but has also shown preclinical activity inducing incomplete autophagy and apoptosis in metastatic cancer models[1][2][3][4][5][6][7].
