Overview

Metoclopramide + granisetron is a combination of two pharmaceutical agents used primarily for the prevention and treatment of nausea and vomiting, particularly in settings such as chemotherapy, radiation therapy, postoperative recovery, and acute migraine. Metoclopramide is a dopamine D2 receptor antagonist that acts centrally on the chemoreceptor trigger zone to prevent nausea and vomiting; it also enhances gastrointestinal motility by increasing gastric emptying. Granisetron is a selective 5-HT3 (serotonin) receptor antagonist that blocks serotonin-mediated signaling in both the central nervous system (area postrema) and peripheral nervous system (gastrocottonintestinal tract), thereby inhibiting emetic pathways. The combination may be considered when single-agent therapy is insufficient or when enhanced antiemetic efficacy is desired.
