This combination therapy is a specific triple-drug regimen consisting of two antibiotics, metronidazole and tetracycline, and a proton pump inhibitor (PPI), omeprazole. It is primarily utilized for the eradication of *Helicobacter pylori* infection, a critical intervention for preventing gastric cancer and treating precancerous gastric lesions. Metronidazole, a nitroimidazole, and tetracycline, a broad-spectrum antibiotic, work synergistically to eliminate the bacteria by disrupting DNA function and inhibiting protein synthesis, respectively. Omeprazole acts by irreversibly inhibiting the H+/K+-ATPase pump in gastric parietal cells, which reduces gastric acid secretion. This acid reduction creates a more favorable pH for antibiotic stability and activity while promoting the healing of the gastric mucosa. This specific combination has been a focus of long-term population-based studies conducted by the Peking University Cancer Hospital & Institute to evaluate the efficacy of *H. pylori* eradication in reducing the incidence and mortality of gastric cancer in high-risk regions.
