Overview

METTL3 siRNA refers to a small interfering RNA molecule specifically designed to silence the expression of the METTL3 gene, which encodes the catalytic core of the RNA methyltransferase complex responsible for N6-methyladenosine (m6A) modification of RNA. Mechanistically, METTL3 siRNA induces sequence-specific degradation of METTL3 mRNA through the RNA interference pathway, leading to decreased METTL3 protein levels and reduced m6A RNA methylation in cells[2][4]. This knockdown results in altered expression and stability of genes regulated by METTL3-dependent methylation, such as NLRP1, KLF4, and ZBTB4, affecting biological processes like angiogenesis, inflammation, cell migration, and atherogenesis[1][2][4]. METTL3 siRNA is primarily used as a research tool for preclinical studies to investigate the role of METTL3 in cancer, vascular, and immune cell biology. There is no evidence it is approved or in clinical trial as a pharmaceutical product.
