Overview

Mevidalen is a small molecule dopaminergic drug developed for the treatment of Lewy body disease, including Parkinson's disease dementia and dementia with Lewy bodies. It acts as a selective positive allosteric modulator (PAM) of the dopamine D1 receptor, enhancing the affinity and activity of dopamine at this receptor. Mevidalen is orally active, crosses the blood–brain barrier, and has been shown to promote wakefulness in sleep-deprived individuals. Its mechanism amplifies responses to endogenous and exogenous dopamine, potentially improving both cognitive and motor symptoms in patients with Parkinson's disease or Lewy body dementias. The drug is being developed by Eli Lilly & Co., with clinical trials reaching Phase 2 for these indications[1][3][4][5].
