Overview

Mevisiran, also known by its code name ENB401, is an siRNA drug developed by EnhancedBio, Inc. It functions as an inhibitor of phosphomevalonate kinase (PMVK), a cytosolic enzyme integral to the mevalonate pathway and isopentenyl diphosphate biosynthesis. The drug is delivered via lipid nanoparticles (LNP). By targeting and inhibiting PMVK, Mevisiran aims to act as a radio-sensitizer, particularly in the treatment of non-small cell lung cancer (NSCLC) and pancreatic cancer. Overexpression of PMVK has been linked to poor prognosis in certain cancers, and its inhibition is expected to enhance the efficacy of chemo- and radio-therapeutic agents, thereby improving patient survival.
