Overview
**Mezigdomide + elotuzumab + dexamethasone** is an investigational three-drug regimen for relapsed/refractory multiple myeloma (MM). Mezigdomide (CC-92480) is a novel, potent oral CELMoD (cereblon E3 ligase modulator) agent that induces degradation of transcription factors Ikaros and Aiolos, leading to apoptosis of myeloma cells and immune stimulation. Elotuzumab is a humanized IgG1 monoclonal antibody targeting SLAMF7 (Signaling Lymphocyte Activation Molecule F7) present on myeloma and natural killer (NK) cells, thereby activating NK cells and facilitating antibody-dependent cellular cytotoxicity against myeloma cells. Dexamethasone is a synthetic corticosteroid with anti-inflammatory and immunosuppressive effects, commonly used as backbone therapy in MM regimens. This combination is currently in clinical trials for patients with relapsed/refractory MM who progressed after anti-CD38 or BCMA-targeted therapies, aiming to provide synergistic immune-mediated cytotoxicity and tumoricidal effects[8][10].
