Overview
**Mezigdomide + trametinib + dexamethasone** is an investigational, all-oral combination therapy studied primarily for relapsed/refractory multiple myeloma. Mezigdomide is a novel cereblon E3 ligase modulator (CELMoD) that induces targeted degradation of transcription factors Ikaros and Aiolos, leading to immune stimulation and anti-myeloma effects. Trametinib is a selective inhibitor of MEK1/2, interfering with the MAPK/ERK signaling pathway critical to cell proliferation, especially in RAS-mutant cancers. Dexamethasone is a synthetic glucocorticoid with anti-inflammatory and cytotoxic properties, frequently used in myeloma therapies. The combination exploits cereblon modulation, MEK inhibition, and steroid-induced apoptosis for enhanced anti-myeloma efficacy.
