Overview

MF59 adjuvanted H5N1 influenza vaccine is a pre-pandemic influenza vaccine that combines H5N1 influenza antigens with the MF59 oil-in-water emulsion adjuvant. MF59 significantly enhances the immunogenicity and efficacy of the H5N1 vaccine by creating a transient "immunocompetent" environment at the injection site. This adjuvant works through multiple mechanisms: it rapidly induces chemokines and inflammatory cytokines, recruits immune cells to the injection site, triggers ATP release from muscle cells, and promotes the differentiation of monocytes into dendritic cells within draining lymph nodes. The vaccine induces stronger hemagglutination inhibiting antibody responses against both homologous and heterologous influenza virus strains compared to non-adjuvanted vaccines, and allows for antigen dose-sparing. Clinical studies have shown that MF59-adjuvanted H5N1 vaccination increases HAI titers by 2 to 5 folds compared to alum-adjuvanted H5N1 influenza subunit vaccine.
