MFA011 is a novel anti-TIGIT and IL-15 fusion antibody developed for cancer immunotherapy, specifically targeting solid tumors. It is an antibody fusion protein that simultaneously acts as an agonist of the interleukin-15 receptor (IL15R) and an inhibitor of T cell immunoglobulin and ITIM domains (TIGIT). The drug is designed to block TIGIT-CD155 interactions, thereby reducing immune suppression in the tumor microenvironment. At the same time, it delivers IL-15 to areas of inflammation, promoting proliferation and activation of CD8+ T cells and natural killer (NK) cells while depleting regulatory T cells (Treg) through antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies have shown that MFA011 increases effector responses leading to enhanced tumor cell killing with good tolerability in animal models[1][2][3].
