MFECP1 is a recombinant antibody-enzyme fusion protein designed for antibody-directed enzyme prodrug therapy (ADEPT). It consists of a high-affinity single-chain Fv (scFv) antibody, MFE-23, which targets the carcinoembryonic antigen (CEA), fused to the bacterial enzyme carboxypeptidase G2 (CPG2). Produced in the yeast *Pichia pastoris*, the protein is mannosylated, which allows for rapid clearance from normal tissues via the mannose receptor in the liver, thereby minimizing systemic toxicity when the prodrug (typically a bis-iodo phenol mustard) is administered. In the ADEPT approach, MFECP1 localizes to CEA-expressing tumor cells, where the CPG2 enzyme converts a subsequently administered non-toxic prodrug into a potent cytotoxic DNA-alkylating agent. This two-stage treatment strategy aims to achieve high concentrations of the active drug specifically within the tumor microenvironment.
