Overview

MFGE8 (Milk Fat Globule-Epidermal Growth Factor 8), also known as lactadherin, is a secreted glycoprotein that has emerged as a potential therapeutic candidate for the treatment of fibrostenosing Crohn's disease. It functions as an anti-fibrotic agent by binding to integrin receptors, specifically integrin αvβ5, which modulates focal adhesion kinase (FAK) signaling pathways to inhibit the production of extracellular matrix (ECM) by intestinal myofibroblasts. Research conducted by institutions such as the Cleveland Clinic and the University of California San Francisco (UCSF) has demonstrated that MFGE8 can prevent and reverse experimental intestinal fibrosis in both in vitro and in vivo models. Its ability to regulate the matrisome in strictured tissues suggests it may offer a novel approach to preventing IBD-induced intestinal strictures and the subsequent need for surgical intervention.
