MG1-hDCT is an oncolytic virus-based cancer vaccine developed by Turnstone Biologics in collaboration with McMaster University. It utilizes a recombinant Maraba virus (strain MG1) engineered to express the human Dopachrome Tautomerase (hDCT) protein, a melanoma-associated antigen. The therapeutic strategy typically involves a heterologous prime-boost regimen: patients are first primed with a replication-deficient adenovirus expressing hDCT (Ad-hDCT) to generate a pool of DCT-specific T cells, followed by a boost with MG1-hDCT. The MG1 virus selectively replicates within tumor cells, leading to direct oncolysis and the release of tumor antigens, while simultaneously acting as a potent adjuvant to expand the DCT-specific T-cell population, thereby enhancing the anti-tumor immune response.
