MG1-MAGEA3 is a genetically modified oncolytic virus-based cancer vaccine. It consists of the attenuated Maraba virus (MG1 strain) engineered to express the human tumor antigen MAGE-A3. The therapy is designed to selectively infect and lyse cancer cells while simultaneously stimulating an anti-tumor immune response by presenting the MAGE-A3 antigen to the immune system. This dual mechanism aims both for direct oncolysis and for induction of robust cellular and humoral immunity against tumors expressing MAGE-A3. The approach has been evaluated as a single agent or in combination with an adenoviral vector encoding MAGE-A3 (AdMAGEA-3) as a priming step, followed by boosting with MG1-MAGEA-3, sometimes in combination with checkpoint inhibitors such as pembrolizumab[2][6][8]. Developed primarily for advanced/metastatic solid tumors expressing MAGE-A3—including non-small cell lung cancer—the drug has shown immunogenicity and safety in preclinical models and early-phase clinical trials[2][5][6].
