Overview

MG1113 is a humanized IgG4 monoclonal antibody that targets the Kunitz-2 domain of tissue factor pathway inhibitor (TFPI), neutralizing both TFPI-α and TFPI-β isoforms. By inhibiting TFPI—a negative regulator of the extrinsic coagulation pathway—MG1113 promotes hemostasis by increasing thrombin generation and rebalancing coagulation in individuals with hemophilia A or B, including those with inhibitors to factor VIII or IX. Preclinical studies have shown that subcutaneous administration of MG1113 restores clotting parameters and reduces blood loss in animal models of hemophilia. The drug is under development for prophylactic treatment of hemophilia and has demonstrated non-linear pharmacokinetics consistent with target-mediated drug disposition.
