Drug intelligence / Profile preview
MG1124 is a highly specific, fully human monoclonal antibody targeting the IgV domain of carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1). Developed by GC Biopharma in collaboration with the Mogam Institute for Biomedical Research, MG1124 binds to the homodimerization interface of CEACAM1, physically disrupting both homophilic and heterophilic inhibitory interactions. This blockade reverses CEACAM1-mediated suppression of T-cell receptor signaling, restoring ZAP70 phosphorylation and interleukin-2 (IL-2) production, and enhances natural killer (NK) cell and CD8+ T-cell cytotoxicity against CEACAM1-expressing tumor cells. In preclinical studies, MG1124 has demonstrated robust tumor growth inhibition in humanized non-small cell lung cancer (NSCLC) patient-derived xenograft (PDX) models, showing strong synergy when combined with PD-1 blockade.
