Overview

MG35-1 is a high-affinity T-cell receptor (TCR) specific for the PRAME (Preferentially Expressed Antigen in Melanoma) protein, presented by HLA-A*02:01. It is the core component of the TCR-T cell therapy candidate **MDG1011**, developed by Medigene AG. The therapy involves engineering a patient's own T cells to express the MG35-1 TCR, which then enables them to recognize and eliminate cancer cells expressing the PRAME antigen. PRAME is a well-validated cancer-testis antigen highly expressed in various hematological malignancies and solid tumors, while having limited expression in healthy tissues. MDG1011 has been investigated in Phase 1/2 clinical trials for the treatment of blood cancers, including acute myeloid leukemia (AML), myelodysplastic syndrome (MDS), and multiple myeloma. Medigene has since shifted its internal focus toward solid tumors but continues to list MDG1011 as a partnering opportunity.
