Overview

MGA shRNA is an experimental short hairpin RNA (shRNA) construct designed to silence the expression of the MGA (MAX-interacting protein) gene. MGA is a transcription factor that functions as a tumor suppressor by antagonizing MYC activity. In preclinical research, particularly in acute myeloid leukemia (AML), MGA shRNA is utilized to model the loss of MGA function, which is frequently observed in various cancers due to inactivating mutations. Silencing MGA with this shRNA has been shown to increase MYC transcriptional activity, promote cell cycle progression through elevated Cyclin E1 and RB phosphorylation, and enhance tumor growth in xenograft models. These constructs are typically delivered via lentiviral vectors to achieve stable knockdown in hematopoietic cell lines.
