Drug intelligence / Profile preview
MGAC-007 is a first-in-class antigen-drug conjugate (AgDC) designed for the treatment of acetylcholine receptor-positive (AChR⁺) myasthenia gravis. Developed by Canopy Immuno-Therapeutics, the biologic is built on a modular platform combining an autoantigen domain, an inhibitory Fc scaffold, and a linker-payload. It operates via a dual mechanism of action: first, it specifically sequesters and clears pathogenic circulating anti-AChR autoantibodies; second, it selectively binds and eliminates autoreactive B-cell receptor (BCR) clones that produce these autoantibodies, while preserving protective immunity. Preclinical studies have demonstrated significant depletion of pathogenic antibodies and selective lysis of AChR-reactive B cells, leading to restored motor function and reduced mortality in animal models.
