Overview

MGB-BP-3 is a first-in-class synthetic polyamide antibiotic, classified as a Strathclyde Minor Groove Binder (S-MGB), developed for the treatment of Gram-positive bacterial infections, particularly Clostridioides difficile-associated disease. Its mechanism of action involves binding to the minor groove of bacterial DNA, interfering with essential processes such as DNA supercoiling and relaxation by inhibiting type II bacterial topoisomerases (gyrase and topoisomerase IV). This results in strong bactericidal activity against Gram-positive bacteria. The drug has completed Phase IIa clinical trials for C. difficile infection and has also been investigated in preclinical studies for skin and soft tissue infections caused by methicillin-resistant Staphylococcus aureus (MRSA). Developed using technology from the University of Strathclyde, MGB-BP-3 represents a novel class of antibacterials with potential applications beyond Gram-positive pathogens[4][5][6][8].
