MGC026 is an investigational antibody-drug conjugate (ADC) developed by MacroGenics for the treatment of advanced solid tumors. It consists of a B7-H3-targeting monoclonal antibody site-specifically conjugated to SYNtecan E, a novel topoisomerase I inhibitor-based linker-payload derived from exatecan, using Synaffix’s GlycoConnect and HydraSpace technologies. The ADC is designed to selectively deliver its cytotoxic payload to B7-H3-expressing tumor cells, which are overexpressed in multiple cancer types but have limited expression in normal tissues. Preclinical studies demonstrated potent antitumor activity across several B7-H3-positive tumor models (including lung, pancreatic, prostate cancers, head and neck squamous cell carcinoma, and melanoma), with favorable tolerability and no observed lung toxicity in non-human primates. Clinical development is ongoing for various advanced solid tumors[1][3][4][5][6].
