MGC028 is an investigational antibody-drug conjugate (ADC) developed by MacroGenics. It targets ADAM9, a member of the ADAM family of multifunctional type 1 transmembrane proteins implicated in tumorigenesis and cancer progression, and is overexpressed in multiple cancers. The ADC consists of an ADAM9-targeting antibody linked to a novel topoisomerase I inhibitor-based cytotoxic payload (SYNtecan E™), utilizing Synaffix’s site-specific linker technology. Preclinical studies have shown that MGC028 exhibits potent, dose-dependent antitumor activity against various solid tumors expressing ADAM9—including gastric, lung, pancreatic, colorectal cancer, head and neck squamous cell carcinoma, cholangiocarcinoma—and has been well tolerated in non-human primates. As of late 2024/early 2025, it has entered Phase 1 clinical trials for advanced solid tumors[1][2][3][4][5][7].
