Overview

MGD-002 is a bispecific Dual-Affinity Re-Targeting (DART) molecule originally developed by MacroGenics and subsequently acquired by Provention Bio (now a subsidiary of Sanofi). It is designed to modulate B-cell activity by co-ligating the inhibitory receptor **CD32B** (FcγRIIb) with **CD79B**, a component of the B-cell receptor (BCR) complex. This co-ligation mimics the natural inhibitory feedback loop where immune complexes dampen B-cell signaling, thereby suppressing B-cell activation, proliferation, and antibody production without causing B-cell depletion. MGD-002 was acquired alongside MGD-010 (PRV-3279), which shares the same targets and mechanism of action. While MGD-010 advanced into clinical trials for systemic lupus erythematosus (SLE), MGD-002 remained a preclinical-stage asset and was not actively progressed into late-stage development.
