MGD-024 is an investigational, next-generation bispecific DART (Dual-Affinity Re-Targeting) molecule that targets both CD123 and CD3. It is designed to engage CD3 on immune effector cells (such as T cells) and redirect them to kill cancer cells expressing CD123, a marker found on various hematological malignancies including acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS). The molecule was engineered to minimize cytokine-release syndrome while maintaining anti-tumor cytolytic activity and allowing for intermittent dosing due to its longer half-life. Its mechanism of action includes antibody-dependent cell-mediated cytotoxicity (ADCC) and T lymphocyte stimulation[1][2][4][5][9].
