MGD007 is a humanized bispecific antibody-based molecular construct known as a Dual-Affinity Re-Targeting (DART®) protein. It is designed to simultaneously bind to the glycoprotein A33 (gpA33), which is highly expressed on colorectal cancer cells, and the CD3 protein found on T cells. By engaging both targets, MGD007 redirects T cells to recognize and eliminate gpA33-expressing tumor cells. Preclinical studies demonstrated potent lysis of gpA33-positive cancer cell lines and inhibition of tumor growth in xenograft models at low doses. Both CD8+ and CD4+ T cells are involved in this process, with activity accompanied by increases in granzyme and perforin expression. The drug was developed for the treatment of relapsed/refractory metastatic colorectal carcinoma.
