MGD015 is a **preclinical bispecific molecule** developed using MacroGenics' proprietary **Dual-Affinity Re-Targeting (DART) platform**. It is designed to simultaneously target **CD3** and an **undisclosed tumor antigen** for the potential treatment of various **hematological malignancies** and **solid tumors**. The molecule works by redirecting T cells, via their CD3 component, to eliminate cancer cells that overexpress the undisclosed antigen. MGD015 has demonstrated the ability to kill targeted cells both in vitro and in vivo, showing high response rates in several mouse tumor xenograft models. Unlike CAR-T cell therapies, MGD015 is manufactured using a conventional antibody platform, providing "off-the-shelf" convenience without requiring genetic modification of T cells from individual patients. The drug was licensed to **Janssen Biotech** in May 2016 under a collaboration agreement worth up to $740 million, with Janssen responsible for clinical development. Janssen indicated plans to initiate a first-in-human study in 2018.
