**MGD020 + MGD014** is a combination of two investigational bispecific DART (Dual-Affinity Re-Targeting) molecules, both developed by MacroGenics, for the treatment of HIV-1 infection in individuals on suppressive antiretroviral therapy. MGD020 targets CD3 on T cells and the gp41 subunit of the HIV-1 envelope, while MGD014 targets CD3 and the gp120 subunit. Each molecule independently binds distinct, highly conserved epitopes of the HIV-1 envelope protein, redirecting CD3+ T lymphocytes to kill HIV-1-infected CD4+ T cells. Used in combination, these agents maximize recognition and cytolytic clearance of infected cells expressing diverse and mutable HIV-1 envelope glycoproteins, aiming to enhance the breadth and potency of anti-HIV activity beyond either agent alone. Preclinical and early-phase clinical studies have shown that the combination mediates potent, consistent redirected T-cell cytotoxicity and antiviral activity in vitro and in vivo, with encouraging safety and pharmacokinetic profiles[1][2][5][7].
