MGD032 is a bispecific DART (Dual-Affinity Re-Targeting) molecule developed by MacroGenics for the treatment of hematological malignancies. It is designed to simultaneously target CD33, a cell surface antigen frequently expressed on myeloid leukemia cells, and CD3, a component of the T-cell receptor complex. By co-engaging these two targets, MGD032 facilitates the recruitment and activation of cytotoxic T cells to specifically eliminate CD33-positive leukemic cells. The molecule was primarily investigated in preclinical models for acute myeloid leukemia (AML), demonstrating potent T-cell mediated killing of CD33+ cell lines and primary AML blasts. While it represented an early candidate in MacroGenics' bispecific pipeline, development appears to have been superseded by other next-generation CD123-targeted or B7-H3-targeted assets.
