Overview

**MGF-E peptide** is a synthetic peptide corresponding to the unique E-domain region (C-terminus) of human mechano-growth factor (MGF), which is an isoform of insulin-like growth factor 1 (IGF-1), specifically the IGF-1Ec splice variant. Unlike mature IGF-1, the MGF-E peptide contains an E-domain generated by alternative gene splicing. This peptide modulates cellular proliferation and has demonstrated activity independent of the IGF-1 receptor. Its principal documented actions include stimulating proliferation and fusion of muscle satellite cells for muscle repair[3], promoting osteoblast proliferation via the MAPK-Erk1/2 pathway[2], displaying neuroprotective effects in ischemia models, and improving cardiac function as well as attenuating pathological remodeling following myocardial infarction in preclinical models[1][5]. The MGF-E peptide operates by a mechanism distinct from canonical IGF-1; its effects are likely IGF-1 receptor-independent, with evidence suggesting roles in nuclear signaling or action at the level of the nucleus, and primary activation of the MAPK-Erk1/2 pathway. There are currently no clinical approvals or well-documented pharmaceutical manufacturers for human use; its use is supported in research applications and preclinical models, primarily for muscle regeneration, bone healing, neuroprotection, and cardiac remodeling.
