Drug intelligence / Profile preview
mgG2 EXCT4 (also known as gG2 EXCT4 or SIM-2) is a preclinical-stage recombinant protein subunit vaccine being developed by Simplexia for the prevention and treatment of genital herpes caused by herpes simplex virus type 2 (HSV-2), as well as associated complications such as neonatal herpes and recurrent (Mollaret's) meningitis. The vaccine candidate consists of a soluble recombinant variant of the normally membrane-bound region of HSV-2 glycoprotein G-2 (mgG-2), designated EXCT4. Unlike other HSV-2 glycoproteins, mgG-2 has low sequence homology with its HSV-1 counterpart, allowing it to induce an HSV-2 type-specific immune response without interference from prior HSV-1 infections. Preclinical studies in mouse models have demonstrated that mgG2 EXCT4 provides robust protection against genital HSV-2 infection and neurological disease.
