Overview

mGlu2 NAM refers to a class of small molecule negative allosteric modulators that selectively inhibit the metabotropic glutamate receptor 2 (mGluR2). Developed by Addex Therapeutics, these compounds are designed to increase glutamatergic tone in the brain by selectively inhibiting presynaptic mGlu2 receptors, which normally act to suppress glutamate release. This selective inhibition is proposed as a novel approach for enhancing cognition without the adverse effects associated with direct activation of ionotropic glutamate receptors. The program is currently focused on mild neurocognitive disorder and has demonstrated proof-of-principle efficacy in preclinical models. The lead compounds are highly selective for mGlu2 and possess good brain penetration[1][2].
