mGlu2 PAM is a small molecule positive allosteric modulator of the metabotropic glutamate receptor 2 (mGlu2) currently in the lead optimization stage at the Warren Center for Neuroscience Drug Discovery (WCNDD). The program is specifically focused on the treatment of Parkinson's Disease. As a positive allosteric modulator, the drug candidate binds to an allosteric site on the mGlu2 receptor, enhancing the receptor's response to its endogenous ligand, glutamate. This mechanism is intended to provide more physiological modulation of glutamatergic signaling compared to orthosteric agonists, potentially offering a better safety profile and reduced receptor desensitization while addressing the neurochemical imbalances associated with Parkinson's Disease.
