mGlu5 NAM refers to a preclinical-stage small molecule program developed by the Warren Center for Neuroscience Drug Discovery (WCNDD) for the treatment of neurodegenerative diseases, including Alzheimer's disease (AD), Parkinson's disease (PD), and Lewy Body Dementia (LBD). The program focuses on the negative allosteric modulation of the metabotropic glutamate receptor 5 (mGlu5). By binding to an allosteric site rather than the orthosteric glutamate binding site, these compounds reduce the receptor's signaling activity in response to glutamate. This mechanism is hypothesized to provide neuroprotective effects by mitigating glutamate excitotoxicity and reducing the aggregation of pathological proteins such as amyloid-beta and alpha-synuclein. The program is currently in the hit-to-lead and lead optimization stages of development.
