MGT-1141 is an antibody-drug conjugate (ADC) designed to target Delta-like ligand 3 (DLL3), a protein highly expressed on the surface of small cell lung cancer (SCLC) and neuroendocrine cancers (NECs) but minimally present in normal tissues. Developed by Marigold, the ADC utilizes a proprietary linker platform to deliver an exatecan payload, which acts as a topoisomerase I inhibitor. Preclinical evaluations have demonstrated that MGT-1141 possesses high target specificity, rapid internalization, and potent anti-tumor efficacy in both cell line-derived and patient-derived xenograft models. Pharmacokinetic and safety studies in cynomolgus monkeys have shown favorable systemic exposure and a wide therapeutic window, supporting its continued development toward clinical investigation.
