MGT-1142 is an exatecan-based antibody-drug conjugate (ADC) engineered to target a novel, tumor-specific glycosylation pattern that is highly expressed in small-cell lung cancer (SCLC) but minimally present in normal tissues. Developed by Mab-Glyco Technology, the agent features an optimized Fc domain for enhanced recognition and selectivity. Upon binding to the target glycan and subsequent internalization, MGT-1142 delivers its potent topoisomerase I inhibitor payload, exatecan, directly into the tumor cells. Preclinical evaluations in cell line-derived and patient-derived xenograft models have demonstrated significant dose-dependent tumor growth inhibition. Pharmacokinetic and safety studies in non-human primates indicate a favorable terminal half-life and a wide therapeutic window, supporting its potential as a first-in-class glycan-targeting ADC for SCLC.
