MGV354 is a novel, selective soluble guanylate cyclase (sGC) activator developed as an ocular therapy for lowering intraocular pressure (IOP), a key modifiable risk factor in glaucoma. It acts by activating sGC, leading to increased cyclic GMP (cGMP) production in human trabecular meshwork cells, which is involved in the regulation of IOP. In preclinical models, topical administration of MGV354 significantly reduced IOP with effects lasting up to 24 hours after a single dose and up to 7 days with repeated dosing. The main adverse effect observed was mild to moderate ocular hyperemia. However, in clinical trials for patients with ocular hypertension or glaucoma, MGV354 did not demonstrate statistically significant efficacy compared to placebo for lowering IOP[1][2][4][5][6][8].
