Overview

MGY825 is an investigational small molecule drug developed by Novartis, currently in first-in-human, phase I clinical trials for the treatment of advanced non-small cell lung cancer (NSCLC). The drug is being evaluated as a single agent, with particular focus on patients whose tumors harbor mutations in the NFE2L2, KEAP1, or CUL3 genes. These genetic alterations are associated with resistance to standard therapies and poor prognosis in NSCLC. MGY825 acts by blocking a specific enzyme involved in cancer cell survival and proliferation; inhibition of this enzyme leads to cancer cell death. The exact molecular target has not been publicly disclosed as of June 2025[1][3][5][9]. The drug is administered orally in capsule form[9].
