MH84 is a small molecule drug candidate described as a novel dual modulator of gamma-secretase and peroxisome proliferator-activated receptor gamma (PPARγ). It is a derivative of pirinixic acid and has been investigated primarily for its potential in treating Alzheimer's disease. Mechanistically, MH84 acts as both a γ-secretase modulator—affecting amyloid precursor protein (APP) processing to reduce beta-amyloid (Aβ1-42) levels—and as an agonist of PPARγ, which may improve mitochondrial function via PGC-1α-dependent pathways. Preclinical studies have shown that MH84 can protect against nitrosative stress, enhance mitochondrial respiration and content, and decrease pathological APP processing in cellular models of Alzheimer's disease[4].
