Drug intelligence / Profile preview
MHA112-rapamycin-tubastatin A is a preclinical, first-in-class, dual-payload antibody-drug conjugate (ADC) designed to promote long-term cardiac allograft acceptance and prevent heart transplant rejection. The ADC consists of MHA112, a monoclonal antibody targeting peripheral node addressin (PNAd) on high endothelial venules (HEVs) in lymph nodes, conjugated to two small-molecule payloads: rapamycin (an mTOR inhibitor) and tubastatin A (a selective HDAC6 inhibitor). By targeting HEVs, the ADC delivers these immunoregulatory payloads directly to lymph nodes, promoting the induction and stability of regulatory T cells (Tregs) and reducing pro-inflammatory T cells. In preclinical murine heart transplant models, MHA112-rapamycin-tubastatin A prolonged graft survival and suppressed chronic rejection at significantly lower doses than the free drugs.
