MHB009C is an investigational antibody-drug conjugate (ADC) developed by Minghui Pharmaceutical for the treatment of advanced solid tumors. It consists of a humanized IgG1 monoclonal antibody targeting B7-H4 (V-set domain-containing T-cell activation inhibitor 1) conjugated to a potent DNA topoisomerase I inhibitor payload via a cleavable linker. The ADC utilizes Minghui's proprietary SuperTopoi platform, which features a payload reported to be significantly more potent than DXd. B7-H4 is an immune checkpoint protein frequently overexpressed in various cancers, including breast, ovarian, and endometrial malignancies, while showing limited expression in normal tissues. Upon binding to B7-H4 on the tumor cell surface, MHB009C is internalized, releasing the cytotoxic payload to induce DNA damage and cell death. It is currently being evaluated in a Phase I/II clinical trial for patients with advanced solid tumors.
