MHES0488A is a monoclonal antibody that targets the Human Epidermal Growth Factor Receptor 2 (HER2/neu). It is characterized by its ability to bind to a distinct epitope on the HER2 protein, which does not overlap with the binding sites of established HER2-targeted therapies like trastuzumab or pertuzumab. MHES0488A was developed as the targeting moiety for the antibody-drug conjugate (ADC) DHES0815A, which incorporates a pyrrolobenzodiazepine mono-alkylator (PBD-MA) payload. In preclinical models of uterine serous carcinoma and other HER2-overexpressing cancers, MHES0488A has been used to evaluate the contribution of the antibody's binding and signaling inhibition relative to the cytotoxic effect of the conjugated ADC. Clinical development of the associated ADC program was conducted by Genentech but was subsequently terminated.
