MI-147 is a small molecule MDM2 inhibitor designed to disrupt the protein-protein interaction between MDM2 and the tumor suppressor p53. Developed as part of a spirooxindole-based series, MI-147 functions by blocking the MDM2 binding pocket, thereby preventing the ubiquitination and subsequent degradation of p53. This restoration of p53 activity triggers cell cycle arrest and apoptosis in cancer cells expressing wild-type p53. MI-147 demonstrated improved cellular potency compared to its predecessor, MI-219, and served as a critical structural intermediate in the optimization process that led to the more potent derivative MI-888.
