MI-463 is a highly potent and orally bioavailable small molecule inhibitor of the menin-MLL interaction. It demonstrates profound on-target activity in MLL leukemia cells, with an IC50 value of 15.3 nM for inhibiting the menin-MLL interaction. MI-463 has shown efficacy in blocking the progression of mixed lineage leukemia (MLL) both in vitro and in vivo, including reducing tumor growth in mouse xenograft models at 35 mg/kg[2][5][6]. The drug is currently for research use only and not approved for clinical use.
