mi-TPPim is a preclinical-stage peptide-based imaging platform developed by Multimmune that targets membrane-bound Heat Shock Protein 70 (mHsp70). While Hsp70 is typically an intracellular chaperone, it is specifically presented on the plasma membrane of a wide variety of human tumors, including head and neck cancer, triple-negative breast cancer, and Barrett's esophagus, but not on normal cells. The mi-TPPim agent consists of a 14-mer tumor-penetrating peptide (TPP) that binds to the oligomerization domain of mHsp70. This peptide can be conjugated to various signaling moieties for multi-modal visualization, including fluorophores (such as IRDye800 or Cy5.5) for fluorescence molecular endoscopy, radioactive isotopes (such as 89Zr via DFO chelators) for PET/CT imaging, and nanoparticles for MRI. It is currently being developed for the detection, visualization, and monitoring of solid tumors.
