MIA-602

**MIA-602** is a synthetic small-molecule antagonist of the growth hormone-releasing hormone receptor (**GHRH-R**). It is part of the "Miami class" of GHRH antagonists. MIA-602 has demonstrated potent **antitumor**, **anti-inflammatory**, and **anti-fibrotic** effects in preclinical models. Mechanistically, it inhibits GHRH-R signaling, resulting in decreased tumor proliferation in a range of cancer cell lines—including acute myeloid leukemia and prostate cancer—by a pathway distinct from conventional chemotherapeutics[1][6][8]. In oncology, it shows efficacy against chemoresistant malignancies. In inflammatory and pulmonary models, including severe COVID-19, MIA-602 reduces inflammatory markers, necroptosis, and tissue injury by modulating pathways such as ZBP1/MLKL necroptosis, NF-κB, STAT3, and MAPK[2][3][5][9]. It also shows beneficial effects on emotional behavior in animal models, likely via anti-inflammatory and antioxidant activity in the brain[7]. MIA-602 is under investigation but is not yet approved for any indication.
