MIAM is a novel indole compound that has demonstrated anticancer activity against human hepatocellular carcinoma (HCC) in both in vitro and in vivo (xenograft) models. Its primary mechanism of action involves the upregulation and activation of Sirtuin-3 (SIRT3), which leads to the induction of apoptosis and cell cycle arrest in the G0/G1 phase. MIAM also influences other cellular pathways, including increasing levels of p53, p21, SOD2, and Bax, while reducing HIF1α and hexokinase II. Furthermore, it activates NADPH oxidase 4 (NOX4) and its subunit p22(phox), and increases cellular reactive oxygen species (ROS). These multifaceted actions suggest MIAM's potential as a therapeutic agent for HCC.
