Overview

Mibampator is a small molecule drug that acts as a positive allosteric modulator (PAM) of the AMPA receptor, an ionotropic glutamate receptor subtype. It was developed by Eli Lilly for the treatment of agitation and aggression in Alzheimer's disease. Mibampator belongs to the biarylpropylsulfonamide group of AMPAR PAMs and is considered a "high-impact" potentiator, capable of robustly increasing AMPAR signaling. In preclinical studies, high-impact AMPAR potentiators like mibampator enhanced cognition and memory at low doses but caused motor coordination disruptions, convulsions, and neurotoxicity at higher doses. Clinical trials in Alzheimer's disease showed no significant cognitive improvement but some benefit on neuropsychiatric measures; development was discontinued after phase II due to lack of efficacy and toxicity concerns[1][3][5].
